Composition:
Artemether 40 mg
Lumefantrine 240 mg
Uses / Indication:
Treatment of acute uncomplicated malaria caused by Plasmodium falciparum and mixed species
Direction of Use:
Adults & children
25-34.9kg: Day 1: 1 tablet initially and 1 tablet again after 8 hours
Days 2 & 3: 1 tablet BID (12 hourly interval)
≥35 kg: Day 1: 2 tablets initially and 2 tablets again after 8 hours
Days 2 & 3: 2 tablets BID (12 hourly interval)
Side Effects:
Common: headache, dizziness, nausea
Less common: QT prolongation, electrolyte imbalance (especially hypokalaemia or hypomagnesemia)
Warnings/Interactions:
Contraindications: Known QT prolongation, serious cardiac arrhythmias.
Interactions:
Avoid grapefruit juice and CYP3A4 modulators (e.g., azoles, rifampin)
May interact with other QT-prolonging drugs (e.g., certain antimalarials, antipsychotics)
Caution:
Monitor in patients with electrolyte disturbances, liver or kidney impairment, or on cardioactive medications
Not recommended in the first trimester of pregnancy unless there is no other alternative treatment available
Therapeutic note: Fat-containing food improves lumefantrine absorption significantly
